In the course of studying the prion-related RNA binding protein TIA-1, a novel mouse model of post-traumatic stress disorder (PTSD) was established. Compared to wild-type controls, TIA-1-deficient mice develop heightened anxiety and avoidance behavior after exposure to contextual fear conditioning, but are otherwise indistinguishable from wild-type littermates in the naive state. These behavioral phenotypes are associated with abnormal stress-dependent synaptic plasticity in the hippocampus. The prion-like properties of TIA-1 were further characterized in budding yeasts. Tia1/Pub1 acts co-operatively with the release factor Sup35/eRF3 to establish a two-protein self-propagating state. This two-protein prion driven by the Q/N-rich prion domains of Sup35 and Tia1/Pub1 can be visualized as distinctive line structures along tubulin cytoskeleton and unveil the role for prion domains in the assembly of multi-protein complexes. |
